Having looked through the resources on Posterous, I'm still a little confused. Which elements do I need to theme, which blocks need to be added etc. Is there a more in depth resource that anyone can point me to, or even better an IRC channel of some sort. I'm pretty good with html and css so that's not a problem for me.

I would suggest taking the default theme (or any simple theme) and picking it apart and adapting until you make it look the way you want. All the pieces you need are in there.
